Gabbar bio Assey Protocol

Requirement:

1- Maize Seed.

2- 15CM petriplates. 3- Germination Paper.

4- Sterile distilled water.

5- Sodium Hypochlorite Solution (3%)

6- Transparent polythene sheets to cover plates.

7- Gabbar test solution. Dilute 2ml Gabbar to 100ml.

8- Gabbar test solution. Dilute 4 ml Gabbar to 100ml.

Method:

1- Soak Maize seed in 3% sodium hypochlorite solution for 5 minute.

2- Wash the seed with 3-5 changes of sterile distilled water.

3- Use sufficient sterile distilled water for seed washing so as to remove even traces of sodium hypochlorite solution.

4- Soak the surface sterilized seeds in sterile distilled water over night for germination.

5- Cut germination paper according to the length of petriplates and Place the paper in each plate.

6-Place the surface sterilized seeds on the petriplates (5-10 seeds per plate for each treatment).

7- Make one replication for each treatment i.e. 50 seeds per treatment.

8- Add 20ml of Gabbar test solution 2% and 4% to respective plates.

9- Add 20ml of distilled water to control as well.

10- Cover the petriplates with transparent polythene sheets to avoid

11- Mark the plates with marker according respective treatment. moisture loss and wait for germination.

12- Allow the maize seeds to grow with plates covered by treatment

transparent polythene sheets.

13- Take observation for root and shoot development after 5-6 days of incubation,

14- Similar protocol can be used for other crops with minor deviation in incubation period for observation of root and shoot development.
